Novartis to Buy Breast Cancer Drug for Up to 3 Billion

Novartis will pay 2 billion dollars upfront for the PI3K inhibitor from Synnovation Therapeutics. The deal aims to bolster its targeted cancer therapy pipeline.

NOVARTIS AG-REG has reached an agreement to acquire a breast cancer drug candidate from the United States-based biotech firm Synnovation Therapeutics for a total consideration of up to $3 billion. The deal involves an upfront payment of $2 billion, with an additional $1 billion contingent on achieving specific development milestones. The experimental drug, SNV4818, is a selective PI3Kα inhibitor designed to treat HR-positive/HER2-negative breast cancer and potentially other solid tumors.

The therapy targets the mutated form of the PI3Kα enzyme, which is frequently implicated in tumor growth, while sparing the normal version of the enzyme found in healthy cells. This selectivity is intended to mitigate the side effects often observed with current PI3Kα-inhibiting treatments. Shreeram Aradhye, the chief medical officer for the pharmaceutical group headquartered in Switzerland, highlighted the clinical hurdles this drug aims to overcome.

While mutated PI3Kα is a well‑established driver in HR+/HER2‑ breast cancer, there remains a challenge in achieving effective pathway inhibition with a tolerable therapeutic profile.

Currently in early-stage trials, SNV4818 has shown encouraging activity against tumors in laboratory settings. The acquisition further expands the oncology pipeline of Novartis, which also includes other targeted treatments such as radioligand therapy candidates. The company anticipates the deal will close during the first half of 2026.
